سایکلین دی۱
سایکلین دی۱ (انگلیسی: Cyclin D1) یک پروتئین است که در انسان توسط ژن «CCND1» کُدگذاری میشود. ژن مذکور بر روی بازوی بلند کروموزوم ۱۱ (باند 11q13) واقع است و در حدود ۱۳٬۳۸۸ جفت باز طول دارد و ۲۹۵ پروتئین را کُدگذاری میکند.[4][5]
اهمیت بالینی
تولید بیش از حد این پروتئین در مراحل آغازین سرطان و پیشرفت تومور مشاهده شدهاست[6] و میتواند از طریق رشد بدون حمایت تکیهگاه سلولی و همچنین رگسازی وابسته به تولید فاکتور رشد اندوتلیال عروقی، منجر به سرطانزایی شود.[7] تولید زیاد این سایکلین دی۱ همچنین سبب کاهش بیان ژنی Fas شده و در نتیجه مقاوت به شیمیدرمانی پیش میآید و سلولهای سرطانی در برابر آپوپتوز محافظت میشوند.[7]
تولید بیش از حد این پروتئین با بقای کمتر بیماران سرطانی و افزایش متاستاز در ارتباط است.[8][9] چنین افزایشی در میزانِ این پروتئین در سرطانهای زیر یافت شدهاست:
- سرطان ریه از نوع سلول غیر کوچک (۳۰–۴۶٪)[10][11]
- سرطانهای بافت پوششی سر و گردن (۳۰–۵۰٪)[12][13][14]
- سرطان لوزالمعده (۲۵٪)[15]
- سرطان مثانه (۱۵٪)[16]
- آدنوم هیپوفیز (۴۹–۵۴٪)[17][18]
- سرطان پستان(۱۳٪)[19][20][21]
بیشتولیدی سایکلین دی۱ بهویژه در «سرطانهای پستان نوع ER+» قویاً تأیید شدهاست[21] و عدم تنظیم آن با بروز مقاومت به هورموندرمانی در این سرطانها مرتبط است.[22][23][24]
جابهجایی کروموزومی جایگاه ژنی سایکلین دی۱ در حدود ۱۵ – ۲۰٪ از موارد مولتیپل میلوما یافت شدهاست.[25][26]
از روشهای رنگآمیزی ایمونوهیستوشیمی آنتیبادیهای سایکلین دی۱ برای تشخیص لنفوم سلول منتل استفاده میشود. همچنین پیشنهاد شده تا از آن بهعنوان یک نشانگر زیستی برای تشخیص سرطان پستان استفاده شود.[27]
